"AI identifies inhibitor for 'undruggable' protein linked to pancreatic cancer"
GIPC1
a naturally occurring human protein whose overexpression in cancer cells drives tumour growth and drug resistance.
PDZ domain
a protein structural region with broad, shallow surfaces that resists binding by conventional small molecules.
Drug discovery has traditionally been a slow, expensive, and largely trial-and-error process. Identifying a molecule that binds to a specific protein target, blocks its harmful activity, and is safe for human use can take over a decade and cost billions of dollars. A central challenge is that many proteins involved in cancer are classified as 'undruggable', their surface geometry or signalling complexity makes it nearly impossible for conventional small molecules to bind them effectively.
Computational approaches to drug discovery have existed since the 1980s, but their power was limited by processing speed and the quality of predictive models. The rise of machine learning and generative AI has transformed this landscape. AI systems can now screen tens of thousands of molecular candidates, predict how they will interact with a target protein, assess toxicity and absorption properties, and narrow the field to a handful of viable candidates, all in a fraction of the time traditional methods require. This convergence of biology, chemistry, and AI is sometimes called computational drug discovery or AI-driven drug design.
India has a growing stake in this space. The country has a strong generic pharmaceutical industry, a large pool of bioinformatics and AI talent, and a significant disease burden from cancers that lack affordable treatments. Collaborations between Indian AI startups and global research institutions represent an emerging model for translating computational capability into clinical impact.
The protein at the centre of this breakthrough is GIPC1 (GAIP-interacting protein, C-terminus 1). GIPC1 is a naturally occurring protein in the human body. It is not inherently harmful. However, when its expression or signalling goes wrong, cancer cells can exploit the molecular pathways associated with GIPC1 to grow uncontrollably or spread to other tissues.
GIPC1 is particularly relevant to pancreatic ductal adenocarcinoma (PDAC), the most common and aggressive form of exocrine pancreatic cancer. PDAC has a five-year survival rate of under 13.3%, driven by late diagnosis, rapid progression, and resistance to chemotherapy. GIPC1 is overproduced in PDAC and actively drives both tumour growth and chemotherapy resistance. Despite this, it had remained undruggable for over a decade.
The reason lies in its PDZ domain, a structural region made up of amino acids, found across bacteria, yeast, plants, viruses, and animals. …

Which of the pairs given above are correctly matched?
Assertion (A): Pancreatic ductal adenocarcinoma (PDAC) has one of the lowest five-year survival rates among common cancers.
Reason (R): PDAC is typically diagnosed at an early stage, but its rapid progression makes treatment difficult.
